What do the terms two-ply, three-ply, four-ply or six-ply mean?
What are the differences between them and how do they affect (the look of) a garment?

Plies refer to twisting of yarns. Two-ply means that two yarns have been twisted together after the initial spinning. In general twisting makes a more durable weave.

Concerning shirtings, a number like 120/2 means that the yarns in both directions have been twisted.
